The core difference between smooth and rough endoplasmic reticulum is that rer has ribosomes on its membrane while ser does not have ribosomes.
The most basic difference between rer and ser is the presence of ribosomes. Rer has ribosomes on the surface but not in ser. Smooth er originates from rough endoplasmic reticulum by giving off the ribosomes while rough er originates from nuclear membrane. While rough endoplasmic reticulum contains ribosomes which are small round organelle have a rough surface and perform protein synthesis.
What is the difference between smooth and rough endoplasmic reticulum. What is smooth er. Smooth er on the other hand helps in the storage of proteins and lipids. Therefore rer is observed as rough while ser as smooth in the microscope.
On the other hand a smooth er does not have ribosomes on its surface. Ser produces lipids and stores them while rer produce proteins and store them as well. Rough er synthesizes and stores proteins. Hence it is called rough er.

Both ser and rer play a vital role in the synthesis and storage of macromolecules. The smooth endoplasmic reticulum is a continuous section of the rough endoplasmic reticulum it just has no or few ribosomes. The main difference between smooth and rough er is that smooth er lacks bound ribosomes whereas rough er is bound with ribosomes.
